1) Caffeine

Caffeine, the most famous nootropic, decreases serotonin and is an agonist to dopamine receptors. Caffeine also increases acetylcholine release by antagonism of local adenosine receptors and enhanced psychostimulation. I advise to start with 200mg and if you’re already used to caffeine, up the dose to 400mg.

2) BCAA + phenylalanine + B6

Serotonin and dopamine are synthesized from tryptophan and tyrosine respectively. BCAA competes with both precursors for uptake in the brain and by just taking BCAA will reduce both serotonin and dopamine. That’s why we combine it with phenylalanine, which is also a dopamine agonist.

Phenylalanine is better than tyrosine at increasing dopamine and phenylalanine also inhibits serotonin synthesis, via inhibiting the enzyme tryptophan hydroxylase 2 (TPH2).

Vitamin B6 is a cofactor in catecholamine synthesis, and B6 supplementation has been shown to boost catecholamines, such as dopamine in the brain. Start with 5g BCAA + 2g phenylalanine and 10mg pyridoxal 5 phosphate (P5P), which is the active form of vitamin B6.

 

3) Creatine

Creatine is very effective at restoring brain bioenergetics at the cellular level. Creatine supplementation reduces plasma free-tryptophan and the free-tryptophan:tyrosine ratio, thus increasing dopamine:serotonin ratio. Creatine also has an antidepressant-like effect which is mediated by an activation of dopamine D₁ and D₂ receptors as well as adenosine receptors. It is best to use 5g.

 

4) Taurine

Taurine is a serotonin antagonist, it increases dopamine and noradrenaline synthesis and is also a GABA agonist. All in all, taurine will calm your mind, as it’s anti-anxiety and a strong GABA agonist, and is well known to enhance mental focus. Use 500-1000mg.

 

5) Forskolin

Forskolin is able to stimulate dopamine formation from tyrosine via tyrosine hydroxylase and also via potassium-evoked stimulation. Forskolin also increases dopamine receptors as well as their sensitivity. Start with 400mg.

6) Magnolia

Magnolia potently lowers cortisol, is antagonistic to serotonin and increases dopamine and acetylcholine release in the brain. Start with 400mg.

7) Epicatechin gallate (EGCG)
